| Summary: | O retículo endoplasmático (RE) desempenha um papel central na síntese, no dobramento e no controle de qualidade de proteínas. Sua homeostase é mantida pelas vias de Resposta às Proteínas Mal Dobradas e de Degradação Associada ao Retículo Endoplasmático, sendo esta última responsável pela retrotranslocação e degradação de proteínas mal conformadas. A Derlin-1 atua como componente-chave do canal de retrotranslocação em associação com a ATPase p97/VCP, sendo ativada por alta demanda proteica. Assim, sua superexpressão tem sido descrita em diferentes tumores, incluindo o câncer de próstata (CaP), no qual se observa forte dependência dos mecanismos de proteostase, sobretudo no CaP resistente à castração (CRPC). De fato, no CRPC há uma intensa reprogramação metabólica e ativação persistente do estresse do RE, de modo que Derlin-1 auxilia na manutenção das células transformadas. Nesse cenário, a modulação farmacológica da Derlin-1 tem se mostrado uma estratégia terapêutica ainda pouco explorada. Compostos naturais são historicamente relevantes, sobretudo na oncologia, e ferramentas de bioinformática estrutural, como modelagem por homologia, docking e dinâmica molecular, aliadas a predições de ADMET, permitem investigar racionalmente sua interação com ligantes. Assim, este referencial teórico situa a Derlin-1 como alvo molecular relevante e explora o composto fenólico 4-nerolidilcatecol como candidato natural bioativo e estruturalmente compatível para abordagens de modelagem e docking molecular voltadas ao CRPC. |
| Abstract: | The endoplasmic reticulum (ER) plays a central role in protein synthesis, folding, and quality control. Its homeostasis is maintained by the Unfolded Protein Response and Endoplasmic Reticulum-Associated Degradation pathways, the latter being responsible for the retrotranslocation and degradation of misfolded proteins. Derlin-1 acts as a key component of the retrotranslocation channel in association with the p97/VCP ATPase, being activated by high protein demand. Accordingly, its overexpression has been described in different tumors, including prostate cancer (PCa), in which a strong dependence on proteostasis mechanisms is observed, especially in castration-resistant prostate cancer (CRPC). Indeed, in CRPC there is intense metabolic reprogramming and persistent activation of ER stress, such that Derlin-1 helps sustain transformed cells. In this scenario, the pharmacological modulation of Derlin-1 has proven to be a still poorly explored therapeutic strategy. Natural compounds are historically relevant, especially in oncology, and structural bioinformatics tools, such as homology modeling, docking, and molecular dynamics, combined with ADMET predictions, allow for the rational investigation of their interaction with ligands. Thus, this theoretical framework positions Derlin-1 as a relevant molecular target and explores the phenolic compound 4-nerolidylcatechol as a bioactive and structurally compatible natural candidate for modeling and molecular docking approaches aimed at CRPC. |
