Abstract: Precision nutrition and the proper balancing of essential amino acids in feed are fundamental to maximizing the feed efficiency of modern broiler strains. Among the essential amino acids, branched-chain amino acids, particularly leucine, play an important metabolic role due to their structural and regulatory functions in skeletal muscle protein synthesis. However, the benefits of leucine supplementation in broilers remain inconsistent in the literature, particularly regarding feed conversion ratio (FCR). Therefore, this study aimed to quantify, through a systematic review and meta-analysis, the effect size of leucine supplementation on the feed conversion ratio of broiler chickens. A systematic search was conducted in the PubMed, SciELO, ScienceDirect, Scopus, and Web of Science databases. Studies comparing broilers fed leucine-supplemented and non-supplemented isonutritive diets with complete dietary descriptions were included. Of the 2,957 studies identified, six met all eligibility criteria and were included in the meta-analysis, comprising a dataset of k = 31 independent comparisons and n = 7,424 broiler chickens. Feed conversion ratio data were analyzed using a random-effects meta-analysis, with the raw mean difference (RMD) between treatment groups used as the effect size measure. Significant heterogeneity was further investigated through meta-regression and subgroup analyses. Overall, leucine supplementation did not significantly affect feed conversion ratio (RMD = 0.002; p = 0.761). However, substantial heterogeneity was detected among studies (I² = 60.97%; p < 0.001). Meta-regression identified nine significant moderators. Subgroup analyses showed that leucine supplementation improved feed conversion ratio in female broilers, during the starter phase (up to 21 days of age), in diets with a leucine-to-lysine ratio ranging from 0.91 to 1.23, and in diets containing more than 190 g/kg of crude protein. Conversely, leucine supplementation impaired feed conversion ratio in male broilers, during the grower phase (21–35 days of age), and when the dietary leucine-to-lysine ratio exceeded 115% of the recommended level. In conclusion, the effects of leucine supplementation on feed conversion ratio are highly dependent on the nutritional context and the sex of the birds.
