| Resumo: | Mayaro virus (MAYV) is the etiological agent of Mayaro fever, a disease characterized by chills, fever, dizziness, gastrointestinal manifestations, headache, ocular pain, myalgia, and arthralgia. Additionally, the infection can progress to a chronic condition in over 50% of patients, characterized by severe and prolonged arthralgia. Nonetheless, there are no specific antiviral drugs or vaccines available against MAYV infection, being the clinical management based on controlling symptoms. In that regard, metal complexes emerge as an approach in drug development due to their therapeutic potential against different viruses. In this study, we evaluated the in vitro antiviral activity of the ligand N-[[4-(phenylmethoxy)phenyl]methyl]-2-pyridinemethanamine (L) and its complexes with palladium (Pd), platinum (Pt), and zinc (Zn) against MAYV. In that regard, Vero E6 cells were infected with MAYV expressing a nanoluciferase reporter (MAYV-nanoluc) and simultaneously treated with the complexes at concentrations ranging from 2.34 to 300 M for 24 h, when virus replication was measured by quantifying nanoluciferase activity. Dichloro-N-[[4-(phenylmethoxy)phenyl]methyl]-2-pyridinemethanamine-Pd(II) [L-Pd(II)] presented the highest SI and was selected for further assays. This complex showed promising activity in inhibiting early stages of MAYV infection, such as entry into the host cells and virucidal effect, suggesting that it may interact with the virus particle. Additionally, L-Pd(II) displayed significant activity on the post-entry stages of the replicative cycle. The data presented here indicates the potential of this complex as a promising candidate for the development of future antiviral drugs against MAYV. |
