Abstract
In this study, we explored the cytotoxic effect of Pollonein-LAAO, an L-amino acid oxidase isolated from the venom of Bothrops moojeni, on prostate cancer cells (PC3), as well as investigating its ability to induce oxidative stress through the generation of reactive species of oxygen (ROS) and effects on intracellular antioxidant systems. The cytotoxicity induced by Pollonein-LAAO at different concentrations (50; 25; 12.5; 6.25; 3.125; 1.56; 0.78; 0.39; 0.19; 0.09; 0.04 μg/mL) after 4 and 24 hours of treatment was analyzed by the MTT assay, and ROS levels were measured by flow cytometry. The activities of antioxidants, such as total glutathione (GSH), glutathione S-transferase (GST), and superoxide dismutase (SOD), were assessed to
understand the role of Pollonein-LAAO in the tumor cell antioxidant system. Pollonein-LAAO at different concentrations significantly reduced the cell viability of PC3 cells (IC50 0.3142 μg/mL) after 24 hours of treatment. Additionally, Pollonein-LAAO (0.19 μg/mL) induced oxidative stress through ROS generation after 4 and 24 hours of treatment. Pollonein-LAAO (0.19 μg/mL) also decreased the level of total GSH and increased SOD activity. The results of this study demonstrate that Pollonein-LAAO induced oxidative stress and altered the activities of antioxidant systems in the PC3 prostate cancer cell line, resulting in cell death. Further
studies are needed to better understand the molecular mechanisms involved in the redox imbalance induced by Pollonein-LAAO.
