Abstract
Coronary Artery Disease (CAD) is characterized by the formation and accumulation of fatty
plaques in the coronary arteries, thus reducing the arrival of oxygen to the heart. Genetic factors
appear to be involved in the development of the disease, such as the M235T variant of the
angiotensinogen gene (AGT) and the insertion/deletion (I/D) polymorphism of the angiotensin converting enzyme (ACE). The objective of this work was to genotype and analyze the
relationship between the M235T variant of AGT and the ACE I/D polymorphism in (CAD).
121 individuals were selected, 74 patients (case) and 47 controls, both undergoing coronary
angiography and analysis of conventional risk factors. Peripheral blood samples were obtained
at the Hospital de Clínicas of the Federal University of Uberlândia (Consent form attached).
and DNA extraction and Polymerase chain reaction (PCR) were carried out to isolate fragments
of the ACE I/D polymorphism and the M235T variant of AGT, the latter being subjected to
enzymatic restriction, with the enzyme Tth111. After these procedures, agarose gel
electrophoresis was performed, stained with ethidium bromide, 3.5% and 2% respectively, for
genotyping of the M235T variant and the ACE I/D polymorphism. The results obtained were
not statistically significant for the M235T variant and the ACE I/D polymorphism in relation
to the occurrence of CAD.
