dc.description.resumoChikungunya virus (CHIKV) is the etiological agent of chikungunya fever, an arboviral disease characterized by symptoms that can become chronic and debilitating. Despite the significant impact of the infection, no specific antivirals are currently available against CHIKV, highlighting the need for novel therapeutic strategies. In this context, extracts from the genus Banisteriopsis, which have already been described as possessing anti-inflammatory and antioxidant activities, may represent a promising source of antiviral compounds. To date, no studies have evaluated Banisteriopsis extracts against CHIKV, making this investigation an innovative approach toward identifying new therapeutic candidates. In this study we performed a dose-response assay to assess the effective concentration of 50% (EC₅₀), cytotoxic concentration of 50% (CC₅₀), and selectivity index (SI) of three extracts from the species Banisteriopsis anisandra, Banisteriopsis argyrophylla, and Banisteriopsis stellaris on CHIKV replicative cycle, using CHIKV-nanoluc, a marked virus carrying a nanoluciferase reporter gene, to infect in Vero E6 cells. Additionally, we applied a time-of-addition assay to elucidate the mechanism of action of each extract. The results indicate that all three extracts possess strong antiviral activity, with EC₅₀ values in the low-hundreds μg/mL range, with no cytotoxicity effects. Consequently, the determined SI for these extracts were higher than 4.6, indicating a favorable antiviral profile. The inhibitory profile was observed across all viral replication stages investigated, abrogating a minimum of 87% of virus replication. In conclusion, this research highlights the potential of Banisteriopsis extracts as a novel anti-CHIKV approach.pt_BR
